I would perform Hektoen Enteric Agar to Isolate the samples for gram negative bacteria. After placing the sample on a hektoen enteric agar only gram negative would grow. The purpose to find why one strain is gram negative is to check if its safe to handle or handle with care because gram negative is more dangerous due to that make treating them more difficult unlike gram positive bacteria.
